Molecular and serological typing of foot-and-mouth disease virus serotypes currently circulating in Egypt

Foot-and-mouth disease (FMD) is an extremely contagious viral affects all cloven- hoofed animals. The present study aimed to investigate the epidemiological situation of FMD in Egypt during 2017 and 2018, based on antigenic genetic characterizations virus (FMDV). Thirty oral epithelia were collected from vaccinated animals (14 native cattle 16 water buffaloes) showed clinical signs four Egyptian governorates having outbreaks. In samples antigen detection was performed using enzyme-linked immunosorbent assay (ELISA), while characterization done by conventional reverse transcription polymerase chain reaction (RT-PCR), Sequencing phylogenetic analysis constructed for characterization. obtained results FMDV ELISA indicated that 50% examined positive serotyped as serotype O 40%, SAT2 33% A 27% respectively. RT-PCR confirmed ELISA. Six amplicons sequenced phylogenetically analyzed protein 1 (VP1) FMD. Results demonstrated genotype related East Africa-3 (EA-3) topotype with 12.7% difference vaccine strain O-IRN-8-2005-Pan-Asia-2. Furthermore, clustered into Asia 6% A-IRN-1-2005. Phylogenetic analysis of VP1 nucleotide sequences demonstrated a close relationship to recent FMD virus isolates from East Africa, rather than to viruses currently circulating in the Middle East.
